Summary
It has been twelve years since Sam went missing. Twelve years of searching and coming up empty each and every time. The Winchester's know who is responsible for the kidnapping: the Yellow-Eyed demon. Everyone has given up on the hope that Sam will return to them.
Everyone but Dean.
Which is why, when a nearby town is discovered barren apart from the bodies of people who had gone missing years before, all children who could be linked back to the demon Azazel, Dean knows that Sam is involved.
One thing Dean knows for sure: he is going to get his brother back, one way or another.
TLDR: Sam was taken by Azazel as a child and has been raised alongside of Azazel's other "children" for many years before reappearing.

Regulus Black had not returned to Hogwarts for his fifth year.
Rumours spread about the castle; with Sirius Black infamously escaping from Grimmauld Place to live with James Potter, the Black Family must have decided Hogwarts wasn't going to change their now only son into what their previous failure had turned into. Some people suppose Regulus has been transferred to another school, others say he was being homeschooled. Whispers within the darkest parts of the castle suggested that Regulus had left his studies for good, to join the rising number of Death Eaters early.
No matter; Regulus had never reappeared, and by the time that the Marauders had graduated from their seventh year, he had been all but forgotten by the students of Hogwarts.
Until the night of November 2, 1980, when he suddenly makes an appearance in the flat shared by Remus Lupin and Sirius Black.
