On Vulcan, biting one's nails is considered a sign of extreme distress and a grievous form of self-harm, so when Spock sees an ensign in sickbay biting her nails, he is appropriately concerned. He learns that it is a human habit, but that does not fully ease his worry, especially when he sees Jim exhibit similar behavior.

“Well I…” he reached for his currently pointed ears.
“Captain, please go.” Spock’s voice was pointed, efficient, cold. “Somehow they do not look aesthetically agreeable on humans.”
Jim saw the way Spock looked at the Romulan commander, felt the tension between them, so it's only natural he thinks Spock might prefer him a different way. That is not, entirely, the case.

Summary
In a Vulcan-Vulcan pair, the first of the two to enter their mating period after a full marriage bond was formed would trigger the mating period of their partner, essentially synchronizing their cycles. Spock had no idea how much, if at all, this would affect Jim, though they both were erring on the side of “not much,” and preparing accordingly.
After all, Jim did not have a mating cycle to trigger.
Jim is starting to think their assumptions might have been just slightly incorrect.
